[pharmacological action]
Pharmacodynamics kanamycin is an aminoglycoside antibiotic. Its antibacterial spectrum is similar to that of streptomycin, but its effect is slightly stronger. It has strong antibacterial activity against most Gram-negative bacteria, such as Escherichia coli, proteus, Salmonella and Pasteurella multocida, and is also sensitive to Staphylococcus aureus and Mycobacterium tuberculosis. Pseudomonas aeruginosa, Gram-positive bacteria (except Staphylococcus aureus), Rickettsia, anaerobic bacteria and fungi are resistant to this product. Similar to streptomycin, sensitive bacteria are susceptible to kanamycin. There was cross resistance with neomycin and unidirectional cross resistance with streptomycin. Escherichia coli and other gram negative bacteria often have acquired drug resistance.
The pharmacokinetics was rapidly absorbed by intramuscular injection, reaching the peak concentration in blood within 0.5-1.5 hours. It was widely distributed in pleural effusion, ascites and parenchymal organs, but rarely infiltrated into saliva, bronchial secretions and normal cerebrospinal fluid. The concentration of drugs in cerebrospinal fluid during meningitis can be doubled. The concentration in bile and feces is very low. It is excreted mainly through glomerular filtration, and 40% -80% of the injection dose is discharged from the urine in its original form. Two hours after a single intramuscular injection of 75mg/kg to goats, the concentration in the urine can reach 8300 μ g/ml. A small amount can be discharged from milk. After intramuscular injection, the elimination half-life of this product in horses, buffaloes, yellow cattle, dairy goats and pigs is 2.1~2.8 hours.
[drug interactions]
(1) It has synergistic effect with penicillins or cephalosporins.
(2) In alkaline environment, the antibacterial effect is enhanced. When combined with alkaline drugs (such as sodium bicarbonate, aminophylline, etc.), the antibacterial effect is enhanced, but the toxicity is also enhanced accordingly. When the pH value exceeded 8.4, the antibacterial effect was weakened.
(3) Combined with cephalosporin, dextran, powerful diuretics (such as furosemide), erythromycin, etc., it can enhance the ototoxicity of this product.
(4) Skeletal muscle relaxants (such as succinylcholine chloride) or drugs with such effects can enhance the neuromuscular blocking effect of these drugs.

[adverse reactions]
(1) Kanamycin has ototoxicity and nephrotoxicity as streptomycin, and its ototoxicity is stronger than streptomycin and gentamicin.
(2) Neuromuscular blockade is often caused by excessive dose.
[precautions]
(1) It has cross allergy with other aminoglycosides, and is prohibited for animals allergic to aminoglycosides.
(2) Use with caution when dehydration or renal function damage occurs to affected animals.
(3) When treating urinary tract infection, oral administration of sodium bicarbonate can enhance the efficacy.
(4) Ca2+, Mg2+, Na+, NH4+, K+and other cations can inhibit the antibacterial activity of the product.
(5) In case of acute poisoning, neostigmine and other anticholinesterase drugs and calcium preparations (calcium gluconate) can be used to antagonize its muscle block effect.
